The microtiter plate provided in ELISA Kit has been precoated with an antibody specific to the target protein. Standards or samples are then added to the appropriate microtiter plate wells with a biotin-conjugated specific polyclonal antibody preparation,and Avidincon jugated to Horseradish Peroxidase（HRP） is added to each microplate well and incubated. Then a TMB substrate solution is added to each well. Only those wells that contain target protein, biotin-conjugated antibody and enzyme-conjugated Avidin will exhibit a change in color. The enzyme-substrate reaction is terminated by the addition of a sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450 nm ± 2 nm. The concentration of the samples is then determined by comparing the O.D. of the samples to the standard curve.

自備物品
1. 酶標儀（建議參考儀器使用說明提前預熱）
2. 微量加液器及吸頭，EP 管
3. 蒸餾水或去離子水，全新濾紙 標本的采集及保存
1. 血清：全血標本請于室溫放置 2 小時或 4℃過夜后于 1000 x g 離心 20 分鐘，取上清即可
檢測，或將標本放于-20℃或-80℃保存，但應避免反復凍融。
2. 血漿：可用 EDTA 或肝素作為抗凝劑，標本采集后 30 分鐘內于 2 - 8° C 1000 x g 離心 15
分鐘，或將標本放于-20℃或-80℃保存，但應避免反復凍融。
3. 細胞培養物上清或其它生物標本：請 1000 x g 離心 20 分鐘，取上清即可檢測，或將標本
放于-20℃或-80℃保存，但應避免反復凍融。
注：以上標本置 4℃保存應小于 1 周，-20℃或-80℃均應密封保存，-20℃不應超過 1 個月，-80℃
不應超過 2 個月；標本溶血會影響zui后檢測結果，因此溶血標本不宜進行此項檢測。
樣本處理：血清或血漿標本*稀釋 10 倍，如：稀釋 10 倍，取 100uL 血清或血漿加入 900uL
樣品稀釋液。建議各實驗室在操作前*行預實驗以建立*稀釋倍數。
