Monosan Mismatch repair （MMR） proteins, MLH-1 highlighted

MMR proteins is a group of nuclear enzymes, which in all proliferating cells participate in repair of base-base mismatch, that occur during DNA replication. In human, 9 genes with MMR function has been identified. Five have particular clinical interest because they may be mutated in families with hereditary non-polyposis colorectal cancer （HNPCC）.
Clone ES05 （MONX11100） was identified as most successful to obtain optimal staining （source: NordiQC）. HIER in alkaline buffer and the use of a 3-step polymer based detection system gave the most robust protocol. Tonsil is a recommendable positive control for MLH1: The mantle zone B-cells must show at least a weak to moderate nuclear staining, while a moderate to strong staining must be seen in the proliferating germinal centre B-cells.
